Abstract

Neurodegenerativediseases(NDDs),includingAlzheimer’sdisease,Parkinson’sdisease,andamyotrophiclateralsclerosis,areprogressivedisorderscharacterizedbyirreversibleneuronallossandcomplex,multifactorialpathologies.Earlyandaccuratediagnosisremainsacriticalchallengeduetooverlappingclinicalsymptomsandtheabsenceofsensitive,disease-specificbiomarkers.Bioinformaticshasemergedasatransformativediscipline,enablingtheintegrationandanalysisofhigh-throughput multi-omics data genomics,transcriptomics,proteomics, metabolomics, and epigenomics alongsideneuroimagingfeaturestouncovernovelmolecularsignaturesanddiagnostictargets.Systemsbiologyandnetwork-basedapproachesfacilitatetheidentificationofdisruptedpathwaysandgene-diseaseassociations,whilemachinelearningandartificialintelligenceenhancepredictivemodelingandpatient stratification.Thisreviewsynthesizescurrentadvancementsinbioinformatics-drivendiagnosticsforNDDs,highlightingtoolsforbiomarkerdiscovery,imaging-omicsintegration,andAI-powereddecisionsupportsystems.Wealsoaddressongoingchallenges,includingdataheterogeneity,standardizationbarriers,andethicalconcerns,andproposestrategicdirectionssuchasreal-timebiosensing,federatedlearning,andunifiedneuroinformaticsplatforms.Collectively,thisreviewunderscoresthepivotalroleofbioinformaticsinrevolutionizingNDDdiagnosisandadvancingthevisionofprecisionneurology.
