>the</segment><segment>aid</segment><segment>of</segment><segment>molecular</segment><segment>technique.</segment><segment>The</segment><segment>genetic</segment><segment>structure</segment><segment>and</segment><segment>conservation</segment><segment>of</segment><segment>various</segment><segment>indigenous</segment><segment>plant</segment><segment>species</segment><segment>from</segment><segment>different</segment><segment>ecological</segment><segment>zones</segment><segment>were</segment><segment>studied.</segment><segment>This</segment><segment>study</segment><segment>explores</segment><segment>genetic</segment><segment>diversity</segment><segment>patterns</segment><segment>inferred</segment><segment>from</segment><segment>molecular</segment><segment>marker</segment><segment>techniques</segment><segment>to</segment><segment>illuminate</segment><segment>population</segment><segment>differentiation</segment><segment>and</segment><segment>ecological</segment><segment>adaptation.</segment><segment>It</segment><segment>also</segment><segment>draws</segment><segment>attention</segment><segment>to</segment><segment>the</segment><segment>need</segment><segment>for</segment><segment>incorporating</segment><segment>novel</segment><segment>genetic</segment><segment>tools</segment><segment>for</segment><segment>biodiversity</segment><segment>assessment</segment><segment>in</segment><segment>less</segment><segment>developed</segment><segment>areas.</segment><segment>The</segment><segment>objective</segment><segment>of</segment><segment>this</segment><segment>study</segment><segment>was</segment><segment>to</segment><segment>evaluate</segment><segment>and</segment><segment>compare</segment><segment>the</segment><segment>genetic</segment><segment>diversity</segment><segment>of</segment><segment>indigenous</segment><segment>plant</segment><segment>populations</segment><segment>from</segment><segment>various</segment><segment>ecological</segment><segment>zones</segment><segment>of</segment><segment>Sindh</segment><segment>using</segment><segment>RAPD</segment><segment>and</segment><segment>SSR</segment><segment>markers.</segment><segment>The</segment><segment>samples</segment><segment>of</segment><segment>leaves</segment><segment>were</segment><segment>taken</segment><segment>from</segment><segment>different</segment><segment>ecozones</segment><segment>coastal,</segment><segment>desert</segment><segment>and</segment><segment>irrigated</segment><segment>zone</segment><segment>of</segment><segment>Sindh.</segment><segment>Genomic</segment><segment>DNA</segment><segment>was</segment><segment>extracted</segment><segment>using</segment><segment>the</segment><segment>CTAB-modiﬁed</segment><segment>method</segment><segment>and</segment><segment>PCR</segment><segment>ampliﬁcation</segment><segment>was</segment><segment>carried</segment><segment>out</segment><segment>using</segment><segment>RAPD</segment><segment>and</segment><segment>SSR</segment><segment>primers.</segment><segment>Agarose</segment><segment>gel</segment><segment>electrophoresis</segment><segment>was</segment><segment>performed</segment><segment>on</segment><segment>amplified</segment><segment>products</segment><segment>and</segment><segment>binary-coded</segment><segment>genetic</segment><segment>data</segment><segment>were</segment><segment>scored</segment><segment>for</segment><segment>statistical</segment><segment>the</segment><segment>use</segment><segment>of</segment><segment>POPGENE,</segment><segment>NTSYS,</segment><segment>and</segment><segment>PAST</segment><segment>software.</segment><segment>High</segment><segment>analysis</segment><segment>with</segment><segment>polymorphism</segment><segment>(78.17%</segment><segment>RAPD;</segment><segment>81.25%</segment><segment>SSR)</segment><segment>indicated</segment><segment>large</segment><segment>genetic</segment><segment>diversity</segment><segment>among</segment><segment>populations</segment><segment>of</segment><segment>the</segment><segment>two</segment><segment>species,</segment><segment>as</segment><segment>shown</segment><segment>from</segment><segment>the</segment><segment>results.</segment><segment>The</segment><segment>highest</segment><segment>genetic</segment><segment>diversity</segment><segment>was</segment><segment>noted</segment><segment>in</segment><segment>desert</segment><segment>regions</segment><segment>such</segment><segment>as</segment><segment>Tharparkar,</segment><segment>and</segment><segment>comparatively</segment><segment>lower</segment><segment>was</segment><segment>observed</segment><segment>in</segment><segment>irrigated</segment><segment>regions.</segment><segment>Cluster</segment><segment>and</segment><segment>PCoA</segment><segment>analyses</segment><segment>further</segment><segment>indicated</segment><segment>that</segment><segment>populations</segment><segment>clustered</segment><segment>according</segment><segment>to</segment><segment>ecological</segment><segment>affin </segment><segment>of</segment><segment>biodiversity,</segment><segment>measuring</segment><segment>the</segment><segment>genetic</segment><segment>composition</segment><segment>of</segment><segment>a</segment><segment>plant</segment><segment>species</segment><segment>on</segment><segment>demand</segment><segment>has</segment><segment>never</segment><segment>been</segment><segment>more</segment><segment>needed.</segment><segment>Genetic</segment><segment>diversity</segment><segment>is</segment><segment>significant</segment><segment>for</segment><segment>its</segment><segment>direct</segment><segment>purpose.</segment><segment>This</segment><segment>has</segment><segment>implications</segment><segment>for</segment><segment>conservation</segment><segment>biology,</segment><segment>and</segment><segment>sustainable</segment><segment>development</segment><segment>(Borah</segment><segment>et</segment><segment>al.,</segment><segment>2021).</segment><segment>Gradient</segment><segment>analysis</segment><segment>of</segment><segment>genetic</segment><segment>So,</segment><segment>variability</segment><segment>within</segment><segment>and</segment><segment>between</segment><segment>plant</segment><segment>populations</segment><segment>is</segment><segment>also</segment><segment>useful</segment><segment>to</segment><segment>identify</segment><segment>unique</segment><segment>genotypes</segment><segment>which</segment><segment>are</segment><segment>under</segment><segment>threat</segment><segment>and</segment><segment>need</segment><segment>immediate</segment><segment>protection.</segment><segment>It</segment><segment>helps</segment><segment>to</segment><segment>devise</segment><segment>new</segment><segment>strategies</segment><segment>for</segment><segment>conservation</segment><segment>too:</segment><segment>each</segment><segment>life</segment><segment>requires</segment><segment>both</segment><segment>in</segment><segment>situ</segment><segment>and</segment><segment>ex</segment><segment>situ</segment><segment>conservation(X.</segment><segment>Zhang</segment><segment>et</segment><segment>al.,</segment><segment>2024).</segment><segment>At</segment><segment>a</segment><segment>larger</segment><segment>scale,</segment><segment>it</segment><segment>is</segment><segment>all</segment><segment>about</segment><segment>genetic</segment><segment>diversity</segment><segment>critical</segment><segment>in</segment><segment>the</segment><segment>enhanced</segment><segment>capacity</segment><segment>of</segment><segment>plants</segment><segment>to</segment><segment>respond</segment><segment>to</segment><segment>evolving</segment><segment>environmental</segment><segment>changes,</segment><segment>climate</segment><segment>included.</segment><segment>change,</segment><segment>pests</segment><segment>and</segment><segment>diseases.</segment><segment>Plant</segment><segment>breeding</segment><segment>programs</segment><segment>also</segment><segment>use</segment><segment>molecular</segment><segment>marker</segment><segment>techniques.</segment><segment>to</segment><segment>identify</segment><segment>those</segment><segment>crops</segment><segment>that</segment><segment>have</segment><segment>the</segment><segment>potential</segment><segment>to</segment><segment>be</segment><segment>resilient</segment><segment>for</segment><segment>negative</segment><segment>biotic</segment><segment>or</segment><segment>abiotic</segment><segment>conditions(Mondini</segment><segment>et</segment><segment>al.,</segment><segment>2009).</segment><segment>As</segment><segment>that</segment><segment>the</segment><segment>use</segment><segment>of</segment><segment>molecular</segment><segment>tools</segment><segment>to</segment><segment>study</segment><segment>biodiversity</segment><segment>gives</segment><segment>not</segment><segment>only</segment><segment>theoretical</segment><segment>knowledge,</segment><segment>but</segment><segment>also</segment><segment>the</segment><segment>potential</segment><segment>to</segment><segment>apply</segment><segment>that</segment><segment>knowledge</segment><segment>in</segment><segment>agriculture</segment><segment>and</segment><segment>environmental</segment><segment>management.</segment><segment>Pakistan</segment><segment>has</segment><segment>been</segment><segment>bestowed</segment><segment>with</segment><segment>diverse</segment><segment>ecological</segment><segment>zones</segment><segment>from</segment><segment>coastal</segment><segment>to</segment><segment>hilly</segment><segment>areas</segment><segment>of</segment><segment>a</segment><segment>variety</segment><segment>of</segment><segment>natural</segment><segment>native</segment><segment>plant</segment><segment>species.</segment><segment>As</segment><segment>such,</segment><segment>this</segment><segment>diversity</segment><segment>is</segment><segment>an</segment><segment>irreplaceable</segment><segment>national</segment><segment>blessing</segment><segment>that<
